EASZY Assay: Estrogen Pathway

Endocrine-disrupting chemicals (EDCs) are widespread in the environment and have been shown to cause a range of harmful effects on reproductive and developmental processes, neurological functions, or the immune system in both human populations and wildlife.

Biobide has validated the OECD TG Nº250 assay using transgenic zebrafish embryos from Tg(cyp19a1b:GFP) with fluorescence in the estrogen receptors. This assay shows high sensitivity, allowing the detection of substances that activate or inactivate estrogen receptors (ER), directly or indirectly, even at low concentrations.

The EASZY Assay is composed of two phases :

1. MAXIMUM TOLERATED CONCENTRATION test is performed to characterize the systemic and developmental toxicity profile. EC10, is calculated for each test substance

2. ESTROGEN RECEPTOR SIGNALING PATHWAY FLUORESCENCE TEST is carried out using the Tg(cyp19a1b:GFP) transgenic zebrafish embryos, expressing green fluorescence protein (GFP) by Estrogen Receptors through the brain aromatase gene promoter (Tong et al., 2009). This model allows for the detection of direct or indirect ER agonists and antagonists. The fluorescence signal is assessed by image analysis and compared to the control mean. A dose-response curve is performed, and potency and hazard indicators are determined with EC50 and statistical differences between groups.

Biobide’s relevant achievements in November

November was definitely an exciting month for Biobide. We received the French Research Tax Credit (CIR) Accreditation, our latest review article was awarded the ACS Editor’s Choice Award and we attended the American Colleague of Toxicology Meeting.

The French Ministry of Higher Education and Research provided Biobide the French Research Tax Credit (CIR) Accreditation. Our French collaborators can now qualify to receive CIR tax incentive through the research and development conducted by Biobide while reducing up to 30% of your eligible expenditure.

This past month also, the scientific review where two of our colleagues (Arantza Muriana and Dr. Iñaki Iturria) collaborated with other entities such as AbbVie and Bristol-Myers Squibb, has been accepted for publication in Chemical Research in Toxicology. Moreover, the review titled “Use of Zebrafish in Drug Discovery Toxicology” was awarded the ACS Editor’s Choice Award on November 16th. You can download the review article in the following link.

Last but not least, once more Biobide did not miss the American Colleague Toxicology (ACT) Annual Meeting. During this 40th Annual Meeting, our USA Business Development Manager, Ane Altuna, presented our latest immunotoxicity and anti-inflammatory studies in zebrafish (download the poster All this great news makes us keep working hard to continue providing the best zebrafish R&D solutions. Good things come in threes!

June has been a very exciting month for Biobide, as we have had the opportunity to attend three major events in USA.

As every year, we started the month attending the largest global event for biotechnology industry, the 2019 BIO International Convention, this year in Philadelphia and represented by ADL Bionatur Solutions the European biotech leader group which Biobide belongs to. Moreover, during the 17-20th we could participate in the Boston 18th Annual World Preclinical Congress (World Pharma Week). This event delivered a cutting-edge agenda, where more than 1500 assitants came together and exchange expertise and opinions in preclinical research and translation across all therapeutic areas. Moreover, Biobide USA’s CEO, Arantza Muriana had the opportunity to deliver our latest presentation, named as ‘Zebrafish Models for Early Research and Development in Neurodegenerative and Neuromuscular Diseases, where modes such as ALS, Duchenne, Dravet Syndrome their application in CNS drug screening and target validation were reviewed.

Last but not least, during the 23-26th, Biobide took part in the 59th Teratology Society Meeting (TSM) in San Diego. The TSM is the premier source for leading research related to birth defects and other disorders of developmental origin. Attendees could gain insight into the causes and prevention of birth defects and disorders of developmental origin. This time, Ms. Muriana, presented Biobide’s latest poster, in collaboration with L’Oreal: ‘Zebrafish and Medaka as Alternative Models for Developmental Toxicity Assessment of Chemicals (Including Behavior as An End Point)’, which is an study focused on the comparison of the potential of both zebrafish and medaka, for the teratogenicity assessment of chemicals.

SETAC EUROPE 2019: New event, new opportunities

The European SETAC annual meeting was held from the 26^th to 30^th of May, coinciding with the 29^th anniversary of the convention. This event gathers large number of professionals from a wide range of fields such as chemistry, toxicology, biology, and ecology. Under the theme “One Environment. One Health. Sustainable Societies”, the major aims of this year’s symposium were the development of practices for the enhancement of sustainable environmental quality and ecosystem integrity. Therefore, focusing attention on the latest methodologies in environmental toxicology. In this way, experts were all convened in the Daughter of the Baltic Sea, better known as Helsinki (Finland).

Naturally, Biobide didn’t miss the opportunity to attend this unique event. In fact, two of our team members, Maitane Ipiñazar (Biobide’s Business Development Manager) and Dr. Iñaki Iturria (Biobide’s Project Manager) were the ones representing the company on this multi-disciplinary meeting. Moreover, we tapped the opportunity to present our two latest posters: ‘Endocrine disruptors screening analyzing zebrafish sperm quality’ and ‘Thyroid disruption screening using zebrafish as a vertebrate model’, and also welcomed the interested to our booth.
