The paper shows in vivo that a single dose of CT1500 enhances re-entrainment to a shifted light dark cycle in a dose-dependent manner in mice and also phase shifts the circadian clock under constant dark with a clear time-of-day related pattern. The phase response curve shows CT1500 causes phase advances during the day and phase delays at dusk. Finally, we show that daily timed administration of CT1500 can entrain the circadian clock to a 24 h rhythm in free-running mice. Collectively, these data support the use of CT1500 in the treatment of disorders of circadian entrainment.

Circadian Therapeutics Limited, a British company focused on the development of therapies to regulate circadian rhythms is pleased to announce the initiation of its first ever clinical programme with patient recruitment starting in their Phase I study for CT-1500 at Nucleus Network’s Melbourne site.

Non-24 Sleep-Wake Disorder is a serious, chronic circadian rhythm disorder very common in people who are totally blind, and it can arise at any age. Currently the treatment options for Non-24, a significant number of whom are blind veterans, are limited. Professor Shantha Rajaratnam, Chair of the Scientific Advisory Board and Deputy Director of the Turner Institute for Brain and Mental Health at Monash University, Australia, commented “ I am delighted that a new clinical programme has started for a promising treatment for Non-24. Circadian Therapeutics Ltd is delighted to announce the appointment of Dr Mark Sampson as Chief Executive Officer and Board Member of the Company, effective 1st July 2020.
Most recently Chief Medical Officer of Shield Therapeutics PLC, Dr Sampson has had a distinguished, 30-year career as a pharmaceutical physician in both Clinical Development and Medical Affairs roles across pharmaceutical, biotech and consumer products companies. He has held leadership roles in medical affairs for firms including Shionogi, Viropharma, Gilead, Amgen and SmithKline Beecham. Dr Sampson began his medical career in the NHS across a range of clinical specialities and holds an MBChB from the University of Glasgow. He was a member of the Prescription Medicines Code of Practice Appeals board for over 10 years.

In the study, NU researchers employed the fruit fly model of Huntington’s disease, a well-studied model organism for both circadian rhythms and neurodegenerative diseases.
The researchers knocked down the hop gene in fruit flies with the protein that causes Huntington’s disease, and were surprised to find that knocking down the gene restored the flies’ induced arrhythmic circadian clocks, reduced the aggregation of diseased proteins in the brain and reduced the number of neurons killed by those proteins.
In the next step, the researchers plan to test this method in a fruit fly model of Alzheimer’s disease. They believe that targeting and knocking down the hop gene could potentially be an early intervention for slowing the progression of various neurodegenerative diseases.

Disturbances to the circadian rhythm, or to the genes that produce the rhythm, can cause problems from low productivity and insomnia to depression and diabetes. Some of the most common disturbances are night-shift work, artificial light and travel across time zones.
At least 10 Israeli research groups focus on chronobiology, the study of biological rhythms. “Compared to Israel’s size we have quite a lot of very diverse research on circadian rhythms,” Bloch tells ISRAEL21c.

Fat cells in the human body have their own internal clocks and exhibit circadian rhythms affecting critical metabolic functions, new research in the journal Scientific Reports, finds.
Researchers led by Dr Jonathan Johnston from the University of Surrey conducted the first ever analysis of circadian rhythms in human fat taken from people isolated from daily environmental changes. Circadian rhythms are approximately 24-hour changes governed by the body’s internal clocks. Misalignment of ‘human clocks’ with each other and the environment is believed to be a major contributor to obesity and poor health.
During this unique study seven participants underwent regulated sleep-wake cycles and meal times before entering the laboratory, where they maintained this routine for a further three days.

Virginia Tech Molecular Biologist Carla Finkielstein’s new research confirms her earlier findings that, this is why night shift workers develop cancer at a higher rate. The bright light they’re exposed to at night confuses cancer suppressing molecules and cancer fueling oncogenes. Finkielstein says, programs limiting how long people stay on that night shift, could help.
The study also finds that the timing of a cancer drug could make all the difference. For example, it may be best to give cancer drugs during the day. That’s when cells are dividing, due to that internal body clock. In cancer, you want to slow down the cells’ divisions, so attacking them while they’re active is most effective.
