Cientistas tiram HIV de 'esconderijo' celular, passo crucial na busca da cura

Um grande obstáculo nas pesquisas em busca da cura para a infecção de HIV é que ele pode se "esconder" dentro de células humanas, adotando uma forma latente que impede sua localização por drogas ou pelo sistema imune. Cientistas anunciaram nesta quarta-feira o sucesso de dois diferentes métodos para "desentocar" o vírus da Aids, abrindo caminho para tentativas de eliminá-lo totalmente do corpo.

Os trabalhos, liderados pela Universidade da Carolina do Norte em Chapel Hill e pela Universidade Emory, de Atlanta, obtiveram sucesso em reativar sinais da presença do vírus em experimentos com macacos e camundongos.

Para tal, os pesquisadores usaram drogas que fazem com que o HIV saia da latência e comece a produzir as proteínas que o compõem, deixando-o exposto.

Um dos experimentos, liderado por Victor Garcia, de Chapel Hill, usou a AZD5588, droga originalmente criada para tratamento de câncer, que também atua numa cadeia de reações imunes.

"Essa abordagem promissora para reversão da latência — em combinação com as ferramentas apropriadas para a liberação sistêmica da infecção por HIV — aumenta muito as oportunidades para liberação", escreveu o cientista com seus coautores em estudo na revista científica Nature, que publica o trabalho.

O outro, liderado por Guido Silvestri, sai na mesma edição, descrevendo uma estratégia diferente. O grupo de Atlanta usou uma combinação de drogas para, ao mesmo tempo, inibir um tipo de célula  imune que cala a atividade do vírus em sua presença (os linfócitos T CD8) e aumenta a produção de interleucina, uma molécula que atua na regulação da imunidade.

Os dois grupos de pesquisa interagiram e colaboraram um com o outro, mas demonstraram cada um deles a eficácia de seus métodos em dois modelos experimentais consagrados de pesquisa. Um deles foi a infecção de macacos resos pelo SIV, o vírus análogo ao HIV que infecta símios. O outro foi a infecção por HIV em "camundongos humanizados", um tipo especial de cobaia.

Como o HIV é capaz de infectar apenas humanos, cientistas alteraram a medula óssea de camundongos, usando tecido humana, para que esses roedores passassem a produzir células vulneráveis ao vírus da Aids.

E tanto nos símios quanto nos roedores, as diferentes estratégias para desentocar o HIV de seu esconderijo funcionaram. Os pesquisadores manifestam esperança de que a estratégia shock and kill (provocar o vírus para depois matá-lo) possa se tornar realidade para humanos. Um passo importante para a transição até testes clínicos é usar antirretrovirais para demonstrar a fase do "kill" (matar) em animais, porque os estudos atuais se dedicaram apenas ao "shock" (assustar).

Apesar de reconhecerem que há um longo de caminho de pesquisa pela frente, cientistas não relacionados com os dois estudos expressaram otimismo. Um desafio a ser enfrentado, ainda, é a habilidade do vírus de plantar seu código genético dentro do DNA. Não está claro ainda se isso pode sabotar as ambições dos pesquisadores na busca da cura, porque o HIV poderia em tese se recriar a partir dos cromossomos humanos. E é preciso saber, também, se há algum grupo de células que resiste às terapias criadas agora para impedir o vírus de se esconder.

"A estratégia shock and kill ainda é em grande medida um conceito teórico, não uma realidade terapêutica", escreveu Mathias Lichterfield, do Hospital Brigham and Women's, de Boston, em um artigo de comentário encomendado pela Nature. Em sua opinião, porém, os trabalhos de Garcia e Silvestri "parecem ser as intervenções mais robustas demonstradas até hoje para perturbar a latência dos vírus".

Hoje, soropositivos conseguem atingir sobrevida de longo prazo, e a infecção já vem sendo descrita por alguns médicos como uma doença crônica tratável. Os antirretrovirais, porém, provocam a resistência do HIV em alguns casos, e em alguns pacientes o organismo não se adapta bem à droga no longo prazo. A busca de uma cura, por isso, ainda é considerada uma meta de pesquisa importante.

Science may have found the secret to a better, and more sustainable, espresso coffee shot - edition.cnn.com

Espresso is one of the most popular ways to drink coffee, but it's also the most complicated to make.
The process depends on a delicate combination of factors -- including grind setting, coffee amount, water pressure, temperature and beverage volume -- and it's hard to consistently pull the perfect shot.

Now, a group of researchers say they've found the key to an espresso that's cheaper, consistently tasty and better for the planet, too. The secret, according to the study published Wednesday in the journal Matter, is to go against traditional espresso wisdom and use fewer coffee beans that are ground more coarsely.

"American companies are catching on to this idea that being able to produce a shot quickly is a positive thing, and being able to produce it reproducibly, of course, is the basis of American consumerisms," said Christopher Hendon, co-senior author of the report and computational chemist at the University of Oregon.
Challenging the norms of espresso

Though a lot of factors are involved, the norm for brewing an espresso is to grind a large amount of coffee beans -- around 20 grams or 0.7 ounces -- as finely as possible.
The fine grind would mean more surface area is exposed to hot water during brewing, which should boost the extraction yield. This is the fraction of the ground coffee that actually dissolves and ends up in the final drink, depending on how water flows under pressure through a mass of coffee grounds.

The Specialty Coffee Association advises that coffee most frequently tastes best when the extraction yield is between 17% to 23%. Coffee beverages exceeding this range typically taste bitter, while those below it are often sour.

But when researchers tested these recommendations, they found the relationship between them was more complicated: grinding as finely as the industry standard clogged the coffee bed, resulting in a reduced extraction yield, wasted coffee grounds and inconsistent flavors.



Finding the perfect recipe

To develop a recipe for consistent extraction yield based on the factors under a barista's control -- the amount of water and coffee, size of the grounds, and water pressure -- the researchers created a mathematical model that helped them narrow it down to two elements: the size of the grind and the amount of coffee used.

"One way to optimize extraction and achieve reproducibility is to grind coarser and use a little less water, while another is to simply reduce the mass of coffee," Hendon said.



Reducing the amount of coffee increases the extraction yield, since the extra room in the coffee bed allows for the water to brew more of the coffee. This means a barista can use 15 grams of coarsely ground coffee and still achieve consistent espresso with the same punch as the standard finely ground 20-gram espresso shot. This modification can result in very fast shots of less than 15 seconds, a higher espresso concentration and a different flavor profile.

Faster shot times happen when brewing coarser grounds because there is more space in the coffee bed for the water to move through.

It's important to note that extraction yield doesn't tell all the qualities of the coffee. For example, coarser grounds can produce a more consistent 23% extraction yield of one flavor, while the finer grounds produce a variation of extractions of different flavors that can average 23%.



"We're not trying to say that all 23 percents taste good," Hendon said. "What we're really trying to say is you need to choose what you think tastes good, and here's a roadmap to sort of navigate back to making that percentage reproducible."

Zachary Gordon-Kane, operations manager of coffee shop and motorcycle lifestyle goods brand Brother Moto coffeeshop in Atlanta, said testing the methods sounds good in practice, but it doesn't take into account perceived body of espresso, or how it will feel in your mouth.

"A lot of people enjoy that espresso can be like a thicker, more viscous experience, with more punch and flavor. When you think about things solely by extraction yield, you can lose a lot of experiences," he said.
Cheaper, and environmentally friendly
Using the methods for espresso production discovered by the authors could also lead to economic benefits for coffee shops and to sustainability benefits for the coffee industry as a whole.



Their model was tested at a local coffeeshop in Oregon using an espresso machine that allowed for control of shot time, water pressure and temperature. By reducing the mass of coffee used by 25%, the cafe was able to save 13 cents per drink, amounting to $3,620 per year.

In addition to the savings, the shot times were reduced to 14 seconds, significantly reducing the order-to-delivery time.



And, being more efficient with coffee bean usage would also reduce waste at a time when coffee supply is under threat from changing climate in historic production areas, the report says.

However, another caveat to the authors' recommendations is that extraction isn't exactly indicative of quality.

While coarser grounds and a shorter shot time produce a consistent result that's just as strong, the traditional method does provide a more complex flavor imparted by partially clogged flow. This is caused by some portions of the grounds undergoing less or more extraction, which can bring out different flavors.
"The upside is that it can taste excellent and complex," Hendon said. "But the downside is that there's natural variation just because of the physics. It's not the barista's fault."

They suggest baristas could blend shots of the same extraction yield to determine which tastes best to them, but this is likely impractical in a busy cafe setting. But if experimenting with espresso is your jam, it's worth a shot.

A Southern restaurant chain known for tiny hamburgers and late-night service has filed for bankruptcy. Federal court documents show the Krystal Co. sought federal bankruptcy protection in Atlanta on Sunday. Documents say the company owes between $50 million and $100 million to food suppliers, equipment companies and others. Krystal was founded in Chattanooga, Tennessee, in 1932 and is now based in Atlanta. With more than 300 restaurants and 7,500 employees, Krystal calls itself the South's oldest fast-food chain. Of Tool Cabinets, Chewing Gum and Sealng Wax #16: Drill Press Table

I’ve been needing a Drill Press Table for quite a while and finally got one built. This is not one of those huge “do everything” tables. It’s 14” deep by 22” wide – mostly made from scrap – which is the reason for the size. I selected/included features that support my methods of work and 90% of the project types I do.

Nothing fancy here, just a couple (2) layers of 3/4” plywood screwed together. I had to clip the tips of the 1-1/2” screws so they wouldn’t poke through.

The table is held to the cast iron table with 5/16” carriage bolts countersunk in to the bottom piece with wing nuts to hold it down.

The table itself has two Rockler T-Tracks dropped into dados made on my table saw using Freud Super Dado set. That Dado Set is really GREAT!! The fit was precise and installation easy.

The edge is trimmed with a few strips of poplar leftover from a library project and have bee sitting around for a long time. The edge does cover the T-Track but the back is open so the T-Bolts can be removed.

The fence is again scrap plywood and poplar. The T-Bolts are actually 1/4-20 hex bolt from the junk drawer and I started with wingnuts but swapped over to 1” knobs I picked up in Atlanta at Peachtree woodworking for $1 each. One if the bolts is capture in a 5/16” hole and the other is in a 5/16” x 2-1/2” slot.

A lot of the DP Table fences I’ve seen have a cut out for the chuck. I decided to just allow the fence to be flipped around to accommodate close-in short, small work.

I wanted a place to set my coffee so on the left side I extended the bottom piece of plywood to create a cup holder/shelf.

Because the table is not really big enough to accommodate a couple of those super cool drawers that some have I decided to put in a swing out box for taps, drills, and miscellaneous stuff.

The box is 5” x 6” x 2-1/2” deep with a small tray a about 1/2” deep

The table size and position allow clearance for both the column release handle

and table elevation crank.

Oh it does have a 3” Square replaceable center and before I set it in place I waxed the cast iron.

Overall I’m really happy with t utility of the table and the features and how it supports my methods of work.

REMEMBERING THE LIFE OF DR. KING

Yesterday we celebrated the life and legacy of Dr. Martin Luther King, Jr. Born in Atlanta, Georgia, on January 15, 1929, Dr. King became the most influential advocate for civil rights in the United States. Through peaceful protests and galvanizing speeches, Dr. King opened the hearts and minds of Americans to no small effect. Up until the moment of his untimely death, Dr. King worked to launch a new era in American society – one that would bring us closer to reaching the ideals set forth by our founding fathers, that “all men are created equal.”

Without anticipating how his words would soon describe his own life, Dr. King once said, “The quality, not the longevity, of one’s life is what is important.” Dr. King was taken from us far too soon, but his legacy lives on forever.

 

PRESIDENTIAL IMPEACHMENT TRIAL BEGINS IN THE SENATE

The articles of impeachment to remove President Donald Trump from office were finally presented to the Senate last week—after nearly a month of delay by Speaker Pelosi. The charges allege the President abused the power of his office by withholding aid to Ukraine in order to pressure the country to begin investigations that could benefit the President politically and that he obstructed Congress by resisting subpoenas issued by the House.

In dramatic fashion, the articles were physically carried from the House through the Capitol to the Senate floor on Wednesday by the seven impeachment managers picked by Speaker Pelosi and formally read aloud. A day later, Chief Justice of the Supreme Court John Roberts was sworn in to preside over the trial, and he swore in the Senators who will serve as the jury, asking them to “do impartial justice according to the Constitution and laws.”  

Opening arguments begin today. It is expected that the trial will closely resemble the impeachment of President Bill Clinton in 1999. Under that precedent, the Senate met every day except Sunday with each side given 24 hours of floor time to present their cases and Senators then given 16 hours of floor time to ask questions. Today, those rules will be offered, and once rules are approved, the trial will officially be underway.

 

PRESIDENT TRUMP SIGNS PHASE ONE TRADE AGREEMENT WITH CHINA; USMCA PASSES THE U.S. SENATE

Last week was significant for U.S. trade policy as President Trump signed the first phase of a trade deal with China. This is an important step forward in President Trump’s efforts to level the playing field with Chinese manufacturers and exporters. The milestone achievement was delivered under the leadership of United States Trade Representative Robert Lighthizer and his team of trade officials as they simultaneously continued working to shepherd the United States-Mexico-Canada Agreement (USMCA) across the finish line in the U.S. Senate. I commend their work, and I want to take this opportunity to share with you some specifics about each of these achievements.

The phase-one trade agreement with China intends to hold Chinese companies and investors accountable for the decades of unfair business practices that they have engaged in at the expense of U.S. workers, farmers, and companies. These practices include forced technology transfers and repressive regulatory schemes, putting U.S. partners at a competitive disadvantage. To combat this, the agreement bolsters American farm exports, opens new markets for American products, curtails China’s longstanding practice of manipulating its currency to unfairly compete against U.S. exporters, and ensures mechanisms are available to effectively implement the agreement. While it’s too early to determine the long-term economic impacts of this trade deal, I am confident that it moves us in the right direction of striking a balance between fostering a successful long-term trade relationship with China while simultaneously making sure our Chinese counterparts play by the rules.

On the heels of President Trump and Chinese Vice Premier Liu signing the first phase of the trade deal with China, the U.S. Senate passed the USMCA on Thursday with overwhelming bipartisan support. President Trump promised during his campaign to renegotiate NAFTA and bring the antiquated agreement into the 21st Century, and the Senate’s approval of the USMCA is a testament to the President’s ability to keep his promise. As I’ve shared with you before, the USMCA is critical to Georgia’s economy. Without it, Georgia’s manufactured goods exports to Canada and Mexico – our two largest export markets – would face between $352 million and $1.5 billion in extra taxes, which would most certainly lead to fewer exports and lower wages or job cuts for Georgians. In 2018 alone, thanks to NAFTA, Georgia exported $10.2 billion worth of goods to Canada and Mexico, and USMCA will help expand that figure. Our poultry industry, which is the largest agriculture industry in the state, is going to find new markets in Canada and Mexico thanks to USMCA. Expanded market access means jobs and investment in our local communities. I look forward to seeing the President sign this measure soon! 

 

EVALUATING FEDERAL EXPENDITURES TO STATES AND LOCALITIES

There is no question that in times of crises – whether natural disaster, economic recession, or act of terror – federal intervention is necessary to provide much needed resources to the regions that are affected. However, in times of an economic upswing as we are currently experiencing, we must reevaluate the federal government’s relationship with states and ask local governments to be better partners in serving our constituencies. States and localities must do more and rely on the federal government less in times of historical prosperity in order for us to focus our resources where they are needed most.

At the House Budget Committee hearing, “Why Federal Investments Matter: Stability from Congress to State Capitals,” held last week, I had a chance to speak with our panel of experts on this very topic. Click on the photo below to watch my questions with witnesses during the hearing.

RESTORING STRENGTH TO ARTICLE ONE

Over the past few decades, Congress has slowly yielded more and more of its power to the executive branch, from deferring to federal agencies to interpret broad legislative language, to shifting resources from the legislative branch to executive. This has come at the expense of Congressional power and has weakened the institution with respect to Article II. I am a firm advocate of Article I as the most accountable and responsive branch of the federal government, and I believe it is best positioned to craft solutions for the American people, which is why it is necessary that we restore our power here in Congress.

During last week’s hearing held by the House Select Committee on the Modernization of Congress, I had a chance to speak with our panel of experts about the different ways we can strengthen Congress. These ideas include brining equity in majority and minority committee resources, building capacity beyond staff numbers, recognizing the utility of bipartisan solutions over nonpartisan recommendations, directing federal spending through Congress rather than the executive, and bolstering Congress’s regulatory ability. CONSTITUENT SPOTLIGHT: EXAMINING U.S. FOREIGN ASSISTANCE

I appreciate hearing the priorities of folks back home concerning U.S. assistance abroad. Below are a few of those messages I have received on this topic.

I urge you to prioritize U.S. leadership by protecting the International Affairs Budget in order to further global poverty reduction efforts, boost U.S. job creation and advance our national security interests.

Please support H.R. 4868, "To certify that United States assistance to the United Nations for humanitarian programs in the Syrian Arab Republic is not misdirected, and for other purposes." The U.S. should not be spending tax dollars for things that do not directly involve the U.S.

According to the most recent report from Global Humanitarian Assistance, an estimated 206.4 million people across 81 countries were deemed in need of humanitarian assistance, with forty percent of those in need residing in only six nations: Yemen, Syria, the Democratic Republic of the Congo, Turkey, North Korea, and Afghanistan.

As Jasmine mentioned, foreign aid is an important tool in pursuing our foreign policy and national defense goals, and I absolutely believe that we have an obligation to help those who cannot help themselves. While there is a robust debate to be had to the extent our nation should be involved in other nation’s affairs and when foreign assistance should be deployed to achieve our shared policy goals, to Larry’s point and support of H.R. 4868, the idea that we should ensure those dollars are being properly managed and used efficiently is a unifying one. Too often we have seen taxpayer dollars in this area mismanaged or diverted from those who they were intended to help. 

With the appropriations process gearing up for Fiscal Year (FY) 2021 and the expected release of the President’s budget request next month, I expect we will revisit the conversation about what areas of the federal budget in this area need continued support and what areas may require restructuring. That conversation is a necessary one, though to be clear, not one rooted in undermining our shared goal of being good neighbors and partners in a global era.

The U.S. has a long and revered history of supporting the humanitarian efforts of others and continues to be one the largest donor of assistance for humanitarian crises around the world. That commitment was surely evidenced in the FY 2020 appropriations package that Congress passed and President Trump signed into law on December 20th. This package provided roughly $9 billion for international security assistance, $1.66 billion for U.S. Agency for International Development (USAID) operations, and a cumulative $8 billion for global health programs. And the good news is we are our seeing our allies collectively increase their contributions in the name of humanitarian assistance.

As Congress begins crafting its appropriations bills for FY2021, I hope to continue to hear about your priorities in this area so we can ensure our nation’s history of goodwill is targeted, effective, and enduring.

 

SIX GWINNETT SCHOOLS RECEIVE DISTINGUISHED TITLE I STATUSES

In the 7th District, we are proud to be continually recognized as having some of the best schools in the country. Last month, the Georgia Department of Education announced its Title I Distinguished Schools. Five Gwinnett County Public Schools elementary schools made the list for the 2019-2020 school year, as well as Buford Middle School from Buford City Schools. The DOE’s Distinguished School designation recognizes the top 5% of the Title I schools in Georgia.

I would like to extend my sincerest congratulations to all these Gwinnett schools. This honor is truly outstanding and a nice reward for all your hard work and dedication.

LOCAL BANDS PLAN BENEFIT CONCERT FOR DESTROYED CUMMING RECORD SHOP

If you ever find yourself knocked down, you can always count on your neighbors in the Seventh District to help lift you up. Two weeks ago, a fire destroyed the entire inventory of His Rock Music, a beloved record store in Cumming where many local bands got their start. A week later, friends of the record store announced they would hold a benefit concert to help restock all that has been lost in the blaze at the Ponce De Leon Music Center on February 8th.

It is such a privilege to live and represent a community that truly takes care of one another. Acts of generosity like these are inspiring and remind us all how special it is to live in this community.

 

LILBURN FAMILY HELPS RAISE $150K FOR DIABETES SUMMER CAMP

The 7th District is always going above and beyond to make our community a better place to live. Last month, Lilburn residents Art and Karen Wood challenged attendees at their Camp Kudzu 20th anniversary celebration to match the fundraising total at last year’s event. But instead of matching last year’s $20,000, attendees raised a total of $150,000! These funds will go to Camp Kudzu, a four-week summer camp designed to educate, empower, and inspire children living with Type 1 diabetes. Last year, the camp had 935 campers from 80 counties across Georgia.

I would like to congratulate Art and Karen for this outstanding fundraising achievement. Your event was a clear success and will go a long way to serve children here in the 7th District and across Georgia.
